How does transcription begin?

RNA polymerase (our train) cannot simply start anywhere on the DNA track. It needs a specific starting point called the promoter - think of it as the train station.

• Promoter sequences are specific DNA regions where RNA polymerase binds to begin transcription
• Transcription factors are helper proteins that assist RNA polymerase in binding to the promoter
• Enhancer sequences can be located upstream or downstream of the gene and boost transcription rates

How can transcription be blocked?

Sometimes cells need to prevent certain genes from being expressed. This is where negative regulatory molecules come in - like a crossing gate blocking the train!

Key Concepts:

• Repressor proteins bind to the operator region of DNA
• When bound, repressors physically block RNA polymerase from passing
• This prevents transcription of downstream genes
• Repressors can be removed when certain signals (like lactose) are present

Why does gene regulation matter?

The regulation of gene expression is directly connected to the observable characteristics (phenotype) of cells and organisms.

Key Concepts:

• Different cells in your body have the same DNA but express different genes
• A muscle cell and a nerve cell look and function differently because of gene regulation
• Environmental factors can influence which genes are turned on or off
• Gene regulation allows organisms to respond and adapt to their environment

What is differential gene expression?

Not all genes are active at all times. Cells selectively express certain genes based on their needs, resulting in different cell products and functions.

• Differential expression means different genes are turned on in different cells or conditions
• This determines what proteins a cell produces
• Cell specialization depends on which genes are expressed
• The same genome can produce hundreds of different cell types

Can RNA regulate genes?

Yes! Beyond carrying genetic information, certain small RNA molecules play important roles in controlling gene expression.

Key Concepts:

• microRNA (miRNA) - small RNAs that can block translation or trigger mRNA degradation
• small interfering RNA (siRNA) - can silence specific genes
• These RNAs work by binding to complementary mRNA sequences
• RNA interference is a natural defense mechanism and research tool
